    Molecular differentiation of infectious pancreatic necrosis virus isolates from farmed and wild salmonids in Ireland

    peer-reviewedThis study investigated the genotypes and sub-groups of infectious pancreatic necrosis virus (IPNV) present in farmed and wild salmonid fish in Ireland. An 1100-bp portion of the VP2 region of segment A from each of 55 IPNV isolates collected over 2003–2007 was amplified by reverse-transcription–polymerase chain reaction and the product directly sequenced. The nucleotide sequences of each isolate were aligned and compared with each other and with the corresponding sequences of a number of reference isolates. All the 55 sequenced isolates belonged to genogroup 5 (Sp serotype) and could be divided into two subgroups. Irish subgroup 1 consisted of isolates from farmed salmon originating from an Irish salmon broodstock. Irish subgroup 2 consisted of isolates from imported farmed stock and all reported clinical outbreaks of IPN were associated with isolates from subgroup 2. Isolates from wild fish were identical to some isolates from subgroup 2, and therefore are believed to have originated from infected farms. These results highlight the importance of import risk analysis for diseases not listed under current legislation

    A second assessment of the whelk fishery Buccinum undatum in the southwest Irish Sea with particular reference to its history of management by size limit

    Whelk landings in the southwest Irish Sea increased from 56 t in 1990 to 6,575 t in 1996 after which they stabilized between 3,600 and 4,600 t annually. At its peak the fishery supported approximately 80 vessels but this number has halved since. In 1994 a size limit of 50 mm was introduced for conservation purposes. Age based assessments of the landings were carried out in 1994, 1996, 1997 and 1999, for which purpose the fishery, ranging from 52º10’ to 53º30’, is divided into four sectors. Landings to the four sectors display biological characteristics which indicate the occurrence of a number of stocklets rather than a single stock unit. Whelk in the south west Irish Sea are relatively thin shelled and the fishery has a low density of large crustacean predators. There is no evidence of contamination by TBT. The northern and southern ends of the fishery have relatively lower densities of larger/older animals; the centre sectors have smaller whelk of shorter life span at higher densities, some of them showing symptoms of a Lee phenomenon and slower growth. A survey of cpue places heaviest densities on the Codling and Rusk Banks, in strong tidal currents, at depths of < 20 m. Growth and maturation rates vary among stocklets. L∞ ranges from 102 to 116 mm. Length at 50% male maturation is usually within the range of 63 – 68 mm and ages of 6.1-7.2 years although landings to one sector have a 50% male maturation rate of 83 mm and 8.5 years of age. The existing size limit of 50 mm would, at best, afford protection to 40% of spawning males. Compliance with the size limit has been poor. From 20 to 33% of total landings in any of the assessed years have been less than the legal limit and in 1994 51% of landings in one sector were below the acceptable size. Trends in cpue have been monitored since 1990 and some areas do not show any marked tendency. On the contrary, some fishermen in the centre sectors improved their yield between 1994 and 1998. There are two explanations for this: the movement of pots onto virgin ground and the fact that fewer fishermen are competing for landings in the same areas. Whelk have responded to a reduction in fishing effort since 1996 immediately following which mortality coefficients (Z) were highest (0.79); they declined to an average 0.61 in 1999. In terms of yield per recruit however they remain high. The southern sector of the fishery is regarded as being most depleted although very few subsize limit whelks were caught there. The survival of the whelk fishery in the southwest Irish Sea is attributed to the instability of the market which is dominated by a single customer, South Korea. A more effective size limit for this fishery would be 68mm (83 mm in the northern sector) and this is considered unrealistic, suggesting that alternative management measures will have to be considered.Funder: Marine Institut

    Indirect Pathogenicity of Haemophilus influenzae and Moraxella catarrhalis in Polymicrobial Otitis Media Occurs via Interspecies Quorum Signaling

    Otitis media (OM) is among the leading diseases of childhood and is caused by opportunists that reside within the nasopharynx, such as Haemophilus influenzae and Moraxella catarrhalis. As with most airway infections, it is now clear that OM infections involve multiple organisms. This study addresses the hypothesis that polymicrobial infection alters the course, severity, and/or treatability of OM disease. The results clearly show that coinfection with H. influenzae and M. catarrhalis promotes the increased resistance of biofilms to antibiotics and host clearance. Using H. influenzae mutants with known biofilm defects, these phenotypes were shown to relate to biofilm maturation and autoinducer-2 (AI-2) quorum signaling. In support of the latter mechanism, chemically synthesized AI-2 (dihydroxypentanedione [DPD]) promoted increased M. catarrhalis biofilm formation and resistance to antibiotics. In the chinchilla infection model of OM, polymicrobial infection promoted M. catarrhalis persistence beyond the levels seen in animals infected with M. catarrhalis alone. Notably, no such enhancement of M. catarrhalis persistence was observed in animals infected with M. catarrhalis and a quorum signaling-deficient H. influenzae luxS mutant strain. We thus conclude that H. influenzae promotes M. catarrhalis persistence within polymicrobial biofilms via interspecies quorum signaling. AI-2 may therefore represent an ideal target for disruption of chronic polymicrobial infections. Moreover, these results strongly imply that successful vaccination against the unencapsulated H. influenzae strains that cause airway infections may also significantly impact chronic M. catarrhalis disease by removing a reservoir of the AI-2 signal that promotes M. catarrhalis persistence within biofilm

    Frequency and significance of post-intubation hypotension during emergency airway management

    Introduction Arterial hypotension is known to follow emergency intubation but the significance of this event is poorly described. We aimed to measure the incidence of post-intubation hypotension (PIH) following emergency intubation and determine its association with in- hospital mortality. Methods A retrospective cohort study of endotracheal intubations performed in a large, urban emergency department over a 1-year period. Patients were included if they were >17 years old and had systolic blood pressure (SBP) >90 mmHg for 30 consecutive minutes prior to intubation. Patients were analyzed in two groups: those with PIH defined by SBP <90 mmHg within 60 minutes of intubation, and those with no PIH. The primary outcome was hospital mortality. Results Emergency intubation was performed on 465 patients, of which 336 met inclusion criteria and were analyzed. The median patient age was 49 years, 59% of patients presented with nontraumatic illness and 92% underwent induction with etomidate. PIH occurred in 76/336 (23%) of patients. The median time to first PIH was 11 minutes (IQR 2 to 27). Intubation for acute respiratory failure was the only independent predictor of PIH (OR = 2.1, 95% CI = 1.1 to 4.0). Patients with PIH had significantly higher in-hospital mortality (33% vs. 21%; 95% CI for 12% difference = 1 to 23%) and longer mean ICU length of stay (9.7 vs. 5.9 days, P <0.01) and hospital length of stay (17.0 vs. 11.4 days, P <0.01). Multivariate logistic regression analysis confirmed PIH as an independent predictor of hospital mortality (OR = 1.9, 95% CI = 1.1 to 3.6). Conclusions PIH occurs in nearly one-quarter of normotensive patients undergoing emergency intubation. Intubation for acute respiratory failure is an independent predictor of PIH. PIH is associated with a significantly higher in-hospital mortality and longer ICU and hospital lengths of stay

    This paper explores first-year undergraduates’ perceptions of the transition from studying geography at pre-university level to studying for a degree. This move is the largest step students make in their education, and the debate about it in the UK has been reignited due to the government’s planned changes to A-level geography. However, missing from most of this debate is an appreciation of the way in which geography students themselves perceive their transition to university. This paper begins to rectify this absence. Using student insights, we show that their main concern is acquiring the higher level skills required for university learning

    Robotic ubiquitous cognitive ecology for smart homes

    Robotic ecologies are networks of heterogeneous robotic devices pervasively embedded in everyday environments, where they cooperate to perform complex tasks. While their potential makes them increasingly popular, one fundamental problem is how to make them both autonomous and adaptive, so as to reduce the amount of preparation, pre-programming and human supervision that they require in real world applications. The project RUBICON develops learning solutions which yield cheaper, adaptive and efficient coordination of robotic ecologies. The approach we pursue builds upon a unique combination of methods from cognitive robotics, machine learning, planning and agent- based control, and wireless sensor networks. This paper illustrates the innovations advanced by RUBICON in each of these fronts before describing how the resulting techniques have been integrated and applied to a smart home scenario. The resulting system is able to provide useful services and pro-actively assist the users in their activities. RUBICON learns through an incremental and progressive approach driven by the feed- back received from its own activities and from the user, while also self-organizing the manner in which it uses available sensors, actuators and other functional components in the process. This paper summarises some of the lessons learned by adopting such an approach and outlines promising directions for future work
